Retatrutide is an investigational, synthetic 39-amino-acid peptide currently being developed for the treatment of obesity, type 2 diabetes, and non-alcoholic fatty liver disease (NAFLD). Developed by Eli Lilly, it represents the next generation of metabolic therapies following dual-agonist medications like tirzepatide.
Mechanism of Action
Unlike earlier single or dual-agonist therapies, retatrutide is a "triple agonist" (triagonist) that simultaneously targets three distinct metabolic pathways:
  • GIP (Glucose-dependent Insulinotropic Polypeptide): Enhances insulin secretion, regulates lipid metabolism, and helps mitigate nausea.
  • GLP-1 (Glucagon-like Peptide-1): Delays gastric emptying, suppresses appetite, and signals satiety in the brain.
  • Glucagon Receptor (GCGR): Increases energy expenditure, stimulates fat burning (lipolysis) in the liver, and directly addresses hepatic fat accumulation. NOT FOR HUMAN CONSUMPTION
